目的研究肝臟低溫保存再灌注期間肝細胞糖原含量與肝細胞凋亡之間的關(guān)系及bax基因在其中的作用。方法制備糖原含量顯著不同的4組兔肝臟模型,根據(jù)給食方法的不同分為A組(禁食24 h,但自由飲水)、B組(標準實驗室飲食)、C組(標準實驗室飲食+每6 h靜脈滴注25%葡萄糖30 ml)及D組(標準實驗室飲食+每4 h靜脈滴注25%葡萄糖30 ml),檢測各組肝臟低溫保存再灌注期間肝細胞凋亡及bax基因蛋白的表達情況。結(jié)果各組肝臟于低溫保存9 h后再灌注60 min時,肝組織內(nèi)可見明顯的肝實質(zhì)細胞凋亡現(xiàn)象,A、B、C、D 4組的凋亡細胞數(shù)量依次減少, 4組間兩兩比較差異有統(tǒng)計學意義,各組肝細胞bax基因蛋白的表達程度與肝細胞糖原含量有密切的相關(guān)性。 結(jié)論肝臟低溫保存再灌注過程中,肝細胞內(nèi)糖原能明顯拮抗肝實質(zhì)細胞凋亡的發(fā)生,其內(nèi)在機理可能為肝細胞糖原通過抑制bax基因蛋白的表達從而達到拮抗肝實質(zhì)細胞凋亡的發(fā)生。
