目的觀察移植胰腺的腺泡細(xì)胞凋亡及其與急性排斥反應(yīng)的關(guān)系。方法選用SD和Wistar大鼠進(jìn)行全胰十二指腸移植。實(shí)驗(yàn)分為同基因移植組(WistarWistar)和異基因移植組(SDWistar)兩組。于術(shù)后第3 d、5 d和7 d分批處死受體,取移植胰腺標(biāo)本用HE染色和原位末端標(biāo)記(TUNEL)技術(shù)檢測(cè)移植胰腺切片,進(jìn)行排斥反應(yīng)的病理學(xué)評(píng)分和計(jì)數(shù)凋亡指數(shù)(AI)。結(jié)果發(fā)生凋亡的細(xì)胞主要是腺泡細(xì)胞,同基因移植組胰腺有散在的腺泡細(xì)胞凋亡,AI在術(shù)后無(wú)明顯變化。異基因移植組胰腺腺泡細(xì)胞凋亡在術(shù)后第3 d、5 d和7 d逐漸升高,AI與急性排斥反應(yīng)的病理學(xué)評(píng)分成正相關(guān)。結(jié)論細(xì)胞凋亡與移植胰腺急性排斥反應(yīng)的嚴(yán)重程度顯著相關(guān),凋亡指數(shù)可作為判斷移植物損傷程度的指標(biāo),對(duì)急性排斥反應(yīng)的診斷有一定的參考價(jià)值。
