對25例大腸癌根治術患者的新鮮標本進行細胞分離和原代培養(yǎng),采用噻唑氮藍(MTT)比色分析法和氚-胸腺嘧啶核苷(3H-TdR)摻入法,在體外觀察5肽胃泌素(PG)對原代培養(yǎng)大腸癌活細胞數(吸光度,Α值)和DNA合成(脈沖數,CPM值)的影響。結果:PG組高、中及低分化腺癌活細胞數和DNA合成均明顯高于對照組(P<0.01,P<0.05)。出現明顯促細胞生長所需PG濃度高分化腺癌組為0.3906μg/ml,而中、低分化腺癌組均為6.2500μg/ml。本研究結果提示:胃泌素可促進人大腸癌細胞的增殖,尤以高分化腺癌對胃泌素更敏感。這為大腸癌患者特別是高分化腺癌應用胃泌素受體拮抗劑進行內分泌治療提供了實驗依據。
