目的 觀察脂多糖(LPS)及前炎癥細(xì)胞因子IL-1β、TNF-α誘導(dǎo)人原代氣道上皮細(xì)胞人β防御素-2(hBD-2)的表達(dá),探討呼吸道先天性病原體防御機(jī)制。方法 LPS、IL-1β及TNF-α刺激培養(yǎng)的人原代氣道上皮細(xì)胞后,RT-PCR法檢測hBD-2 mRNA的表達(dá),Western blot和免疫組化法檢測hBD-2蛋白的表達(dá)。結(jié)果 正常人原代上皮細(xì)胞有微量hBD-2 mRNA表達(dá),不同質(zhì)量濃度的LPS、IL-1β、TNF-α刺激細(xì)胞后,hBD-2 mRNA表達(dá)呈劑量依賴性增加,與對照組差異顯著。0.1 μg/mL的LPS、1 ng/mL的IL-1β和10 ng/mL的TNF-α刺激上皮細(xì)胞4 h后,細(xì)胞均可見hBD-2蛋白表達(dá)。結(jié)論 一定劑量的LPS及前炎性細(xì)胞因子可誘導(dǎo)人氣道上皮細(xì)胞hBD-2表達(dá),且hBD-2可較快表達(dá),hBD-2的表達(dá)可能是氣道最初的防御反應(yīng)。
