目的  探討IL-10 對(duì)小鼠巨噬細(xì)胞髓樣分化因子88( MyD88) / 核因子κB( NF-κB) 炎癥信號(hào)活化的影響。方法  將小鼠巨噬細(xì)胞Ana-1 分為脂多糖( LPS) 組和LPS + IL-10 組, 分別于0. 5、1及2 h 收集巨噬細(xì)胞和細(xì)胞培養(yǎng)上清液,Western blot 檢測(cè)細(xì)胞MyD88 與胞漿、胞核NF-κB p65 亞基表達(dá), ELISA 法檢測(cè)培養(yǎng)上清中腫瘤壞死因子α( TNF-α) 含量。結(jié)果  在0 ~2 h, LPS 組細(xì)胞MyD88表達(dá)顯著持續(xù)上升, LPS +IL-10 組于LPS刺激后上升, 0. 5 h 達(dá)峰值, 2 h 恢復(fù)至正常水平, 1 h 和2 h相對(duì)含量均低于LPS 組( 11. 6 ±1. 3 比17. 5 ±0. 7, 8. 8 ±0. 3 比21. 4 ±1. 8, P  lt; 0. 05) ; 總NF-κB 表達(dá)量在兩組間無明顯差異。NF-κB 核漿比變化趨勢(shì)與MyD88 類似, LPS + IL-10 組1 h 及2 h 相對(duì)含量亦均低于LPS 組( 1. 1 ±0. 1 比2. 4 ±0. 4,0. 6 ±0. 7 比3. 1 ±0. 6, P  lt;0. 05) ; 相應(yīng)的, LPS+ IL-10 組1 h和2 h TNF-α含量亦低于LPS 組[ ( 222. 5 ±33. 5) pg/mL 比( 365. 2 ±22. 7 ) pg/mL, ( 212. 7 ±15. 9) pg/mL比( 566. 2 ±31. 5) pg/mL, P  lt;0. 05] 。結(jié)論  IL-10 通過抑制減少M(fèi)yD88/NF-κB 信號(hào)通路活化, 降低TNF-α表達(dá), 從而下調(diào)炎癥反應(yīng)強(qiáng)度。
