目的 探討抗腫瘤相關(guān)糖蛋白-72(TAG-72)單鏈抗體的原核表達及與4種肝癌細胞系和肝癌組織的特異性親和力。
方法 將抗TAG-72單鏈抗體的cDNA片段插入噬菌粒載體pCANTAB5E, 獲得噬菌粒載體TAG-72-scFv-pCANTAB5E。將后者轉(zhuǎn)化入E.coli HB2151, 經(jīng)β, D異丙基硫代半乳糖苷(IPTG)誘導表達獲得抗TAG-72單鏈抗體蛋白。采用細胞培養(yǎng)及免疫細胞/組織化學方法, 檢測4種肝癌細胞系及石蠟包埋的肝癌組織中TAG-72抗原的表達。
結(jié)果 SDS-PAGE及Western blot法證實, 抗TAG-72單鏈抗體蛋白分子得以正確表達?？筎AG-72單鏈抗體可與肝癌細胞系SMMC7721、HepG2和HHCC結(jié)合, 表明這3種細胞表達了特異性腫瘤抗原; 抗TAG-72單鏈抗體不能結(jié)合BEL7402細胞。40例肝癌組織中TAG-72抗原表達陽性率Ⅰ期、Ⅱ~Ⅲ期分別為23.08%(3/13)和62.96%(17/27),在正常肝組織標本中無TAG-72抗原表達,TAG-72抗原在正常肝組織及肝癌組織中表達的差異有統(tǒng)計學意義(P lt;0.05)。
結(jié)論 TAG-72抗原在肝癌細胞或肝癌組織中有特異性表達，有可能成為判斷肝癌的標志物。
